Arriving to Bari airport, you can take a bus or a train to Bari Centrale train station and then a train to Lecce, where you can take a direct bus to Otranto.

Next take a train from Bari to Lecce, costs from 10 to 19 Euros one way depending on the train you take and takes around 1 hr 30 minutes, www.trenitalia.com.
